Microdroplet Emulsion Generator Arrays for High-throughput Single Cell Genetic Variation Analysis

چکیده

Single cell resolution technology enables the investigation of rare biological events masked by the ensemble average. We have developed a high-throughput microdroplet emulsion generator array (MEGA) as a core platform for massively parallel emulsion PCR-based detection of single cells with a frequency lower than 10 [1, 2]. We have recently used this platform to detect E. coli O157 in a background of non-pathogenic bacteria [2]. Now, we extend this system to screening human genomic DNA (gDNA) for the t(14;18) translocation, a genetic marker associated with lymphoma, as well as extracting single mammalian cell genomes for analysis.
